ABSTRACT

Different physical and chemical stimuli are detected by the peripheral sensory receptors of dorsal root ganglion (DRG) neurons, and the generated inputs are transmitted via afferent fibers into the central nervous system. The gene expression profiles of DRG neurons contribute to the generation, transmission, and regulation of various somatosensory signals. Recently, the single-cell transcriptomes, cell types, and functional annotations of somatosensory neurons have been studied. In this review, we introduce our classification of DRG neurons based on single-cell RNA-sequencing and functional analyses, and discuss the technical approaches. Moreover, studies on the molecular and cellular mechanisms underlying somatic sensations are discussed.

ABSTRACT

Objective@#To discuss the functional recovery after recurrent laryngeal nerve injury (RLNI) on different electromyography thresholds during thyroid surgery.@*Methods@#The prospective experimentally were induced in 12 acute recurrent laryngeal traction animals (porcine) from December 2014 to December 2015, the amplitude and latency of electromyography, even time course during RLNI and recovery of 24 recurrent laryngeal nerves(RLN) were continuous intraoperative neuromonitoring(IONM), including 12 RLN releasing traction after 50% amplitude decrease (AD) and other 12 RLN after 70% AD. The IONM data and postoperative laryngoscopy result of 1 119 thyroid cancer patients, involved 237 male and 882 female, aged 45.2 years in average, who underwent thyroidectomy in Department of Thyroid Surgery, China-Japan Union Hospital Affiliated to Jilin University from July to December in 2016 were analyzed retrospectively.@*Results@#The porcine model of traction lesion showed that the time of 50% AD was (59±4) s, latency increase (LI) was (8± 4)%, was recovered in 10 minutes; the time of 70% AD was (75±6)s, LI was (11±5)% , was recovered (43±23)% of baseline even during 20 minutes. Among the IONM of 1 632 recurrent laryngeal nerves in clinic, the mechanism of 64 RLNI is clear, including traction injury accounted for 62.5% (40/64), thermal injury was 12.5% (8/64), compression injury was 23.4% (15/64), clamp injury was 1.6% (1/64). When 50%≤AD <70% (8.8%), the rate of abnormal vocal fold movement (AVCM) was 8.8% (6/68), while AD≥70% (37.2%), the rate of AVCM was 37.2% (19/51), but LOS was 5/13.@*Conclusion@#Releasing the injury before AD≥50% in surgery, is a more effective indicator to avoid postoperative AVCM and promote nerve function recovery.

ABSTRACT

Stereotactic body radiotherapy (SBRT) can treat the lesions in asurgical manner and inhibit tumor cell proliferation.Furthermore,it has a character of non-invasive,which shortens the duration of treatment.Although surgery and radiotherapy are the major means for early stage peripheral non-small cell lung cancer,some studies in recent years show that SBRT has certain advantages in local control ratio and adverse reaction compared with radiotherapy,and acquires the similar curative effect with surgery.

ABSTRACT

<p><b>BACKGROUND</b>It has been proved that lysosome-associated protein transmembrane 4 beta (LAPTM4B) plays an important role in tumorigenesis, the *2/*2 genotype of LAPTM4B was closely related to liver cancer susceptibity. The aim of this study is to investigate the possible association between the allelic variation of LAPTM4B and the genetic susceptibility of lung cancer.</p><p><b>METHODS</b>The genotype of LAPTM4B was detected in 131 patients with lung cancer and 104 unrelated healthy adult individuals as control by polymerase chain reaction based on special primers. The genotypic distribution of LAPTM4B was analyzed by Chi-square test.</p><p><b>RESULTS</b>The allelic frequencies of the *1 and *2 were 74.8% and 25.2% in the lung cancer group and 72.1% and 27.9% in the healthy control group respectively, but no significant difference was observed between the two groups (Chi-square=0.433, P=0.510). The genotypic distribution of the *1/*1,*1/*2 and *2/*2 were 53.4%, 42.7% and 3.8% in the lung cancer group, 54.8%, 34.6% and 10.6% in the healthy control group respectively, there was no significant difference between the two groups (Chi-square=4.89, P=0.087). No remarkable association was observed between the genotypic distribution of LAPTM4B and the clinical information in the patients with lung cancer, including pathological type and TNM staging.</p><p><b>CONCLUSIONS</b>The results suggest that the allele of LAPTM4B is not closely associated with genetic susceptibility of lung cancer.</p>

ABSTRACT

<p><b>BACKGROUND</b>The present experimental data have showed that the function of kinase suppressor of Ras (KSR) is mainly as a scaffold protein that coordinates the assembly of a multiprotein complex containing MAPK and its upstream regulators. But whether KSR has kinase activity is still the point of argument until now. The aim of this study is to construct eukaryotic expression vectors of carboxyl terminus and amino terminus of KSR and to detect their expression in 293T cell line.</p><p><b>METHODS</b>N-KSR and C-KSR were amplified by PCR. The eukaryotic expression vectors of pCMV-Tag2b-N-KSR and pCMV-Tag2b-C-KSR were constructed by gene recombination technique and the recombinant plasmids were verified by restriction enzyme analysis and sequencing. Then positive clones were transfected into 293T cell line. Expression of target proteins was analyzed by Western blot.</p><p><b>RESULTS</b>The sequences and open read frames of the two vectors were both completely concordant with experiment design. The target proteins could be observed in transfected 293T cells by Western blot.</p><p><b>CONCLUSIONS</b>Eukaryotic expression vectors of pCMV-Tag2b-N-KSR and pCMV-Tag2b-C-KSR are successfully constructed, and they can be expressed in 293T cells. It provides an experimental base for further research work.</p>

ABSTRACT

<p><b>BACKGROUND</b>Multiple drug resistance is one of the important reasons that patients failed to chemotherapy and died of tumors. The aim of this study is to investigate the expression of resistance-related proteins, P-glycoprotein (P-gp), multidrug resistance-associated protein (MRP), lung resistance-related protein (LRP), topoisomerase II (Topo II) and glutathion-S-transferase-π (GST-π) in non-small cell lung cancer (NSCLC) tissues and their clinical significance.</p><p><b>METHODS</b>Expression of P-gp, MRP, LRP, Topo II and GST-π was measured in 98 NSCLC tissues by immunohistochemistry SP method.</p><p><b>RESULTS</b>The positive rate of P-gp, MRP, LRP, Topo II and GST-π was 27.6%, 11.2%, 75.5%, 50.0% and 64.3% respectively. Significant relationship was observed between expression of LRP and TopoII and histological classification. No remarkable difference in expression of multiple drug resistance-related proteins was found among different TNM stages. A total of 47 patients with NSCLC were treated with chemotherapy. There was significant relationship between expression of P-gp, LRP and GST-π and chemotherapy response (P < 0.05).</p><p><b>CONCLUSIONS</b>NSCLC has overexpression of multiple drug resistance-related proteins. Combined detection of these proteins may be useful to guide chemotherapy and predict the response for lung cancer.</p>

ABSTRACT

Objective:To explore the influence of liquid for post-operative irrigation on the effect of ESS.Method:192 cases with chronic sinusitis and/or nasal polyps of groupIIstageII who had undergone the ESS from September,1998 to June,1999 and completed a3-month follow-up were divided randomly into 3 groups to accept 3 different kinds of irrigation liquid.The evaluation of curative effect was according to the index of HaiKou ESS-97.Result:The curative effect of Herba Houttuyniae group was better than the other two.Conclusion:The post-operative care is important and the irrigation liquid can affect the effect of ESS.

ABSTRACT

Angiogenesis-related diseases involving EGF include acherosclerotic plaques, haemangioma, angiofibroma, tumor growth and arthritis. EGF may serve as a drug target and its antagonists may have important clinical applications.Peptide phage display libraries have been successfully applied in areas of finding ligands for enzymes, receptors, and many other molecules. A pⅧ-based peptide phage display library was panned with the cytokine EGF and several EGF-binding clones were selected based on ELISA and micropanning assays. The selected EGF-binders from peptide phage display library may be utilized in affinity chromatography in EGF downstream processing and even act as potential antagonists of EGF if their affinity is further improved through secondary library strategy.
